Receptor level activity follows the canonical GHRH pathway. The peptide binds the GHRH receptor, a class B G protein coupled receptor on somatotroph cells of the anterior pituitary. Binding raises intracellular cyclic AMP, which promotes calcium entry and the release of growth hormone into the bloodstream. Because the molecule acts at the same receptor as the endogenous hormone, its effect is superimposed on the natural pulsatile rhythm rather than replacing it. Whether sustained receptor occupation leads to desensitization is not fully settled.
CJC-1295 belongs to a class of synthetic peptides modeled on growth hormone releasing hormone, a forty-four amino acid signal produced by the hypothalamus. The compound is built from the first twenty-nine residues of the natural sequence, with four substitutions introduced at positions 2, 8, 15 and 27. These changes were designed to slow enzymatic breakdown while preserving receptor activation. The result is a peptide that is shorter than native GHRH and considerably more resistant to ordinary clearance pathways in circulation.
Two related forms appear under the CJC-1295 name. The simpler analogue, commonly written as modified GRF(1-29), carries the four substitutions but no additional conjugation. The second form attaches a maleimidopropionic acid linker to a lysine residue, a modification frequently called the drug affinity complex. That linker reacts with the free thiol on serum albumin to form a covalent bond. Because albumin has a long residence time in blood, the conjugated peptide stays in circulation far longer than the unconjugated analogue.
Downstream of growth hormone, the liver and other tissues increase production of insulin-like growth factor 1, a mediator of many growth-promoting effects. Studies have documented elevated levels of both hormones after dosing, and the rise from the long-acting form persists longer than that produced by shorter-acting analogues. What remains unclear is whether sustained elevation of these markers translates into meaningful clinical benefit, and whether prolonged exposure carries risks that short trials could not detect.
CJC-1295 acts at the growth hormone-releasing hormone receptor, a G-protein-coupled receptor found on somatotroph cells in the anterior pituitary. Binding triggers a rise in cyclic AMP and calcium entry, which promotes release of stored growth hormone. Because the peptide mimics the body's own releasing hormone, it amplifies existing secretory pulses rather than driving continuous output. The size of the response therefore depends partly on the subject's own hormonal rhythm and feedback state.

The drug affinity complex is a maleimidopropionic acid group attached to a lysine side chain. It reacts with the free thiol of cysteine-34 on circulating albumin, forming a covalent bond. This conjugation keeps the peptide in the bloodstream and shields it from rapid renal filtration and proteolysis. Reported circulation half-lives for the albumin-bound form fall in the range of roughly six to nine days in early human studies.
Binding to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and stimulates growth hormone synthesis and release. Because the peptide acts upstream of the pituitary, effects are mediated through endogenous growth hormone rather than direct receptor activation in peripheral tissues. Increases in insulin-like growth factor 1 are generally described as a downstream consequence. Most published human exposure data come from small early-stage studies, and the clinical significance of the pharmacokinetic profile remains incompletely characterized.
CJC-1295 is a synthetic analogue of growth hormone-releasing hormone, built on the 29-amino-acid GHRH(1-29) fragment. Four substitutions distinguish it from the natural sequence: D-alanine at position 2, glutamine at position 8, alanine at position 15, and leucine at position 27. These changes reduce enzymatic cleavage and extend the peptide's persistence in circulation. The compound is discussed in two forms, one carrying a drug affinity complex and one without it.
The peptide backbone includes a D-alanine at position two, which resists cleavage by dipeptidyl peptidase IV, and several other substitutions that reduce degradation. Its molecular weight is roughly 3.4 kDa without the linker and about 3.6 kDa with it. The molecule is water soluble and is normally supplied as a lyophilized powder. Precise sequence and mass values depend on which variant is described, so technical documents usually state the exact form being referenced.
CJC-1295 is a synthetic peptide designed as a long-acting analogue of growth hormone-releasing hormone (GHRH). Its structure derives from the first 29 amino acids of native GHRH, a fragment often called GRF(1-29). Four substitutions were introduced to slow enzymatic breakdown and extend activity relative to the natural sequence. The compound was developed by ConjuChem as part of a broader effort to improve the pharmacokinetic profile of peptide hormones. It is studied in laboratory and clinical research settings rather than appearing as a naturally occurring substance.
== Mechanism of action == In addition to Bcl-2, navitoclax also inhibits the related Bcl-xL and Bcl-w proteins. Because navitoclax inhibits Bcl-xL, it reduces platelet lifespan, causing thrombocytopenia, and this makes it dose-limiting. A 2020 article published in Aging Cell details the synthesis of Nav-Gal (navitoclax-galactose), a novel prodrug via galacto-conjugation of navitoclax and its superiority. The prodrug reduces thrombocytopenia in treated mice at therapeutically effective doses, as well as apoptosis of platelets in human blood samples treated ex vivo. Nav-Gal is efficient for selective senolysis and is passively taken up by both non-senescent and senescent cells.
The Cold War was reflected in culture through music, movies, books, television, and other media, as well as sports, social beliefs, and behavior. Major elements of the Cold War included the perceived threat of communist expansion, a nuclear war, and – connected to both – espionage. Many works use the Cold War as a backdrop or directly take part in a fictional conflict between the United States and the Soviet Union and their respective allies. The period 1953–62 saw Cold War themes becoming mainstream as a public preoccupation.
The authors of the 2007 Comprehensive Assessment of Water Management in Agriculture cited poor governance as one reason for some forms of water scarcity. Water governance is the set of formal and informal processes through which decisions related to water management are made. Good water governance is primarily about knowing what processes work best in a particular physical and socioeconomic context. Mistakes have sometimes been made by trying to apply 'blueprints' that work in the developed world to developing world locations and contexts. The Mekong river is one example; a review by the International Water Management Institute of policies in six countries that rely on the Mekong river for water found that thorough and transparent cost-benefit analyses and environmental impact assessments were rarely undertaken. They also discovered that Cambodia's draft water law was much more complex than it needed to be. In 2004, the UK charity WaterAid reported that a child dies every 15 seconds from easily preventable water-related diseases, which are often tied to a lack of adequate sanitation. Since 2003, the UN World Water Development Report, produced by the UNESCO World Water Assessment Programme, has provided decision-makers with tools for developing sustainable water policies. The 2023 report states that two billion people (26% of the population) do not have access to drinking water and 3.6 billion (46%) lack access to safely managed sanitation. People in urban areas (2.4 billion) will face water scarcity by 2050.

Natural GnRH was previously prescribed as gonadorelin hydrochloride (Factrel) and gonadorelin diacetate tetrahydrate (Cystorelin) for use in treating human and dairy cattle diseases respectively. Modifications of the decapeptide structure of GnRH to increase half life have led to GnRH1 analog medications that either stimulate (GnRH1 agonists) or suppress (GnRH antagonists) the gonadotropins. These synthetic analogs have replaced the natural hormone in clinical use. Its analogue leuprorelin is used for continuous infusion, to treat breast cancer, endometriosis, prostate cancer, and following research in the 1980s by researchers, including Dr. Florence Comite of Yale University, it was used to treat precocious puberty. The expression of GnRH receptors in cancers has led to the use of GnRH as a targeting molecule to deliver toxins specifically to the receptor-expressing cancer cells. In a similar concept, its use to deliver toxins to pituitary gonadotropes in animals has been explored as a means of sterilization, with limited success. GnRH was also shown to successfully deliver DNA into the pituitary gonadotropes where the expressed protein blocked expression of the hormones that regulate reproduction. A Cochrane Review is available which investigates whether GnRH analogues, given before or alongside chemotherapy, could prevent damage to women's ovaries caused by chemotherapy. GnRH agonists appear to be effective in protecting the ovaries during chemotherapy, in terms of menstruation recovery or maintenance, premature ovarian failure and ovulation.

==== Systemic criminality ==== It is often said that all or most users sustain their addiction via crime, however academic Mark Hunter shows that the majority work for extremely low pay, often for other poor people.
In May–June 1897, Thomson investigated whether or not the rays could be deflected by an electric field. Previous experimenters had failed to observe this, but Thomson believed their experiments were flawed because their tubes contained too much gas. Thomson constructed a Crookes tube with a better vacuum. At the start of the tube was the cathode from which the rays projected. The rays were sharpened to a beam by two metal slits – the first of these slits doubled as the anode, the second was connected to the earth. The beam then passed between two parallel aluminium plates, which produced an electric field between them when they were connected to a battery. The end of the tube was a large sphere where the beam would impact on the glass, created a glowing patch. Thomson pasted a scale to the surface of this sphere to measure the deflection of the beam. Any electron beam would collide with some residual gas atoms within the Crookes tube, thereby ionizing them and producing electrons and ions in the tube (space charge); in previous experiments this space charge electrically screened the externally applied electric field. However, in Thomson's Crookes tube the density of residual atoms was so low that the space charge from the electrons and ions was insufficient to electrically screen the externally applied electric field, which permitted Thomson to successfully observe electrical deflection.

August 17, 2009: Japan The Japanese Government informed that after fifteen continuous months of shrinking, and after the most disastrous fall since World War II of 4% between January and March, its GDP grew 3.7% in the second quarter of 2009 in comparison with the same quarter of 2008. In annual terms, Japanese GDP grew 0.9% between April and June. Recovery was mostly contributed to recovering demand in the manufacturing sector with exports growing 6.3%. Consumer spending had only increased by 0.8% in the same period.

== Economic role == These markets are less dependent on imported goods than supermarkets due to their smaller volumes and lesser emphasis on consistency. They have been described in a 2019 food security study as "critical for ensuring urban food security", particularly in Chinese cities. Their roles in supporting urban food security include food pricing and physical accessibility. Academic papers in urban studies, studies on food distribution, and the Singapore National Environment Agency have noted lower prices, greater freshness of food, and the facilitation of both bargaining and social interaction as key reasons for the persistence of these markets. Their persistence has also been attributed to "culinary traditions that call for freshly slaughtered meat and fish as opposed to frozen meats". In developing countries with agriculture-based economies, fresh meat is mainly distributed through traditional markets or meat stalls. Markets selling fresh meat are often attached to, or located near, slaughter facilities.
After optimizing the concentration of graphene oxide concentration and anisotropic porous structure, tensile tests were performed. It was found that MGCA could recover 99.8% and 96.3% when compressed to 60% and 90% strain, respectively. SEM images showed that due to its unique structure, MCGA pore walls were able to wrinkle and fold during compression. Another unique characteristic of this material is its absorption capacity of 80-197 times its weight towards hydrophobic compounds, such as oils and chemical solvents. On the contrary, less effort has been made to study the stretchability of porous carbons. Gao, et al. synthesized a long-range lamellar scaffold composed of chitosan and graphene oxide via bidirectional freezing, freeze drying, and annealing. The result is a material with density of 11 mg cm−3 and porosity of about 99.4%. Various tensile tests were conducted, and it was found that carbon spring could revert to its original shape upon 80% compression strain and -60% stretching strain with a Poisson's ratio between 0.05 and 0.1. The narrow hysteresis loop of the stress-strain curve indicates a low energy dissipation (energy loss coefficient of about 0.2) because of its negligible interior friction, localized buckling, or cracks during deformation processes. The stretchable mechanical properties of this material allow for great candidates for vibrational and magnetism sensors.
Insulin shock therapy or insulin coma therapy was a form of psychiatric treatment in which patients were repeatedly injected with large doses of insulin in order to produce daily comas over several weeks. It was introduced in 1927 by Austrian-American psychiatrist Manfred Sakel and used extensively in the 1940s and 1950s, mainly for schizophrenia, before falling out of favour and being replaced by neuroleptic drugs in the 1960s. It was one of a number of physical treatments introduced into psychiatry in the first four decades of the 20th century. These included the convulsive therapies (cardiazol/metrazol therapy and electroconvulsive therapy), deep sleep therapy, and psychosurgery. Insulin coma therapy and the convulsive therapies are collectively known as the shock therapies.
The relationship between sex steroids and SHBG in physiological and pathological conditions is complex, as various factors may influence the levels of plasma SHBG, affecting bioavailability of testosterone.
==== Absorption ==== Minoxidil is readily absorbed from the gastrointestinal tract with oral administration. Its absorption from the gut is around 90% or more. The drug reaches peak levels after about 30 to 60 minutes. Following attainment of peak levels, concentrations of minoxidil rapidly decline. Sublingual minoxidil is expected to have higher bioavailability than topical minoxidil. Peak levels with sublingual administration occurred after 30 minutes. The bioavailability of oral minoxidil is not affected by food and it can be taken in either a fasted or fed state. In the case of topical administration to the scalp, the absorption of minoxidil is only about 1.2 to 1.4%. With this route, serum levels of minoxidil are usually less than 5 ng/mL and are frequently undetectable. It has been predicted that application of 5% topical minoxidil twice to the entire scalp might be equivalent to a single 5.4 mg oral dose of minoxidil in terms of systemic exposure. The stratum corneum of the scalp is saturated by minoxidil and acts as a reservoir for the drug. This results in a continuous flow of minoxidil in the scalp, with absorption being completed after about 10 to 12 hours. Based on these findings, topical minoxidil is generally applied twice daily. A wet scalp has been found to increase the absorption of topical minoxidil. Peak levels of minoxidil with oral minoxidil were 16.8 ng/mL with 2.5 mg, 37.2 ng/mL with 5 mg, and 74.7 ng/mL with 10 mg doses. Mean peak minoxidil levels with a single 0.45 mg dose of sublingual minoxidil were 1.62 ng/mL (range 0.3–5.3 ng/mL).

The names are often used interchangeably in casual writing, but they are not strictly identical. Modified GRF(1-29) refers to the unconjugated analogue carrying only the four substitutions. The version sold as CJC-1295 with DAC includes the albumin binding linker and clears much more slowly.
DAC stands for drug affinity complex, a term describing the maleimidopropionic acid linker. This group forms a covalent bond with the cysteine residue on serum albumin after administration. The resulting adduct is what extends the circulating half life from minutes to days.
No. It is entirely synthetic and does not correspond to any known endogenous peptide. The design starts from human growth hormone releasing hormone and introduces deliberate substitutions. Any biological effect comes from mimicking the natural hormone at its receptor.
It binds the growth hormone-releasing hormone receptor on pituitary somatotroph cells. Receptor activation raises cyclic AMP and promotes release of stored growth hormone granules. Because the peptide persists longer than natural releasing hormone, stimulation is prolonged rather than brief.
